L-tryptophan is a protein building block your diet has to supply, while 5-HTP is a compound your body makes from tryptophan on the way to serotonin.

The two ingredients sit in separate bottles on our shelf, except for one melatonin capsule that holds both, and that capsule is the easiest place to double up without noticing.

Where do tryptophan and 5-HTP sit in the serotonin pathway?

Tryptophan comes first and 5-HTP second: your body converts tryptophan into 5-HTP, and an enzyme then converts 5-HTP into serotonin.

MedlinePlus Genetics lays out the chain on its page about the DDC gene. Tryptophan is converted to 5-hydroxytryptophan, then an enzyme called aromatic L-amino acid decarboxylase (AADC) removes a carboxyl group, producing serotonin. The page describes serotonin as a chemical messenger that carries signals between nerve cells and other cells.

The chain keeps going. NCCIH's overview of sleep and complementary health approaches groups L-tryptophan and 5-HTP as substances your body can change into melatonin.

So a 5-HTP supplement joins the chain one step later than tryptophan does. That's a map of the route, not a promise. We don't claim either supplement raises serotonin, and none of our labels says so.

Tryptophan also has jobs 5-HTP doesn't. It's part of the protein your body builds, and according to the niacin fact sheet from the NIH Office of Dietary Supplements (ODS), your liver converts a share of it into niacin (vitamin B3).

Is 5-HTP an amino acid?

Not in the usual sense: 5-HTP is a modified form of the amino acid tryptophan, but it isn't one of the protein-building amino acids MedlinePlus lists, and it isn't essential.

The full name, 5-hydroxytryptophan, tells the story. It's tryptophan with a hydroxyl group (one oxygen, one hydrogen) attached at position 5 of the molecule. That small change makes it a different compound.

The MedlinePlus entry on amino acids defines amino acids as molecules that combine to form proteins, and it names tryptophan among the nine essential ones. 5-HTP doesn't appear on the page.

In MedlinePlus's terms, essential means the body can't make it, so it has to come from food. ODS describes tryptophan as an amino acid in proteins and singles out turkey as high in it.

Is 5-HTP or L-tryptophan better for sleep?

Neither 5-HTP nor L-tryptophan has earned the title of better sleep supplement. NCCIH says both have sedative effects, but how they work is unclear and the data are very limited.

That's NCCIH's plain verdict, and we'd rather pass it on than dress up either one. The same NCCIH page notes that a 2017 sleep-medicine guideline advised against tryptophan for long-running sleep trouble.

Our labels reflect the gap. The 5-HTP Capsules' directions tie the dose to a meal and never mention bedtime. The two products carrying L-tryptophan are sleep formulas that lean on melatonin too, per 2-capsule serving: 2 mg in the valerian capsule and 10 mg in the 10mg Melatonin Sleep Capsules.

So if you're shopping by the word "sleep," the real question isn't 5-HTP or tryptophan. It's which whole formula fits, and whether you want melatonin in it.

Which is safer, 5-HTP or L-tryptophan?

No federal page we found calls 5-HTP or L-tryptophan the safer pick, and NCCIH's firmest warning, about serious reactions alongside serotonin medicines, names L-tryptophan.

In our words, NCCIH says L-tryptophan supplements may cause a dangerous, even life-threatening, serotonin reaction when combined with medicines that change how the body handles serotonin. Anyone on such a medicine needs the prescriber's approval first.

Tryptophan also carries a separate, older safety story, about contamination rather than drug combinations. NCCIH says supplement use of L-tryptophan stopped in 1989, when an illness outbreak was traced to one company's contaminated synthetic product. The page doesn't say when it returned, but L-tryptophan is back on labels today, ours included.

NCCIH adds that several 5-HTP sources, tested because of the close chemical tie to tryptophan, showed no similar contaminants. That's a purity result, not a sign that either ingredient is easier on you, so it doesn't settle this comparison.

How much 5-HTP and tryptophan do our labels list?

Per 2-capsule serving, our 5-HTP Capsules list 200 mg of 5-HTP, the valerian capsule lists 100 mg of L-tryptophan, and the 10 mg melatonin capsule's blend lists both, unmeasured.

The takeaway: only the melatonin capsule carries both, but any two of these products together stack the same chain twice.

Each of those 5-HTP and tryptophan entries carries a footnote saying the Daily Value isn't established, so nothing on the panel says 200 mg is a lot or 100 mg is a little. Don't read 200 against 100 as "twice as strong," either, for reasons the FAQ below covers.

The blend needs a little math. Under federal labeling rules, a blend names its ingredients from heaviest to lightest (we unpack that rule in what a proprietary blend is). This blend names 14 ingredients. If the panel follows that rule, L-tryptophan, in first place, weighs at least one-fourteenth of 905 mg, about 64.6 mg. 5-HTP, in last place, can't weigh more than that same one-fourteenth.

The combinations we'd flag, per serving of each:

  • 5-HTP Capsules plus the valerian capsule: 200 mg of 5-HTP and 100 mg of L-tryptophan, two steps of one chain.
  • 5-HTP Capsules plus the 10 mg capsule: 5-HTP from two bottles, one in an unknown amount, next to the blend's St. John's wort.
  • The valerian capsule plus the 10 mg capsule: at least 164.6 mg of L-tryptophan if the ordering rule holds, plus 12 mg of melatonin.

NCCIH says taking St. John's wort with medicines that affect serotonin may lead to serotonin-related side effects, which can be serious.   2. Valerian & Chamomile Sleep Capsules. The label's suggested use is two capsules daily for adults, with 6 to 8 oz (177 to 237 ml) of water, at no set time.
  3. 10mg Melatonin Sleep Capsules. The label's suggested use is two capsules once a day, 20 to 30 minutes before bedtime, with an 8 oz (237 ml) glass of water.

Every one of these labels warns, "Do not exceed recommended dose." Stick to one serving of one product, and don't layer a second tryptophan or 5-HTP product on top.

Label details can settle it. The 5-HTP capsule's shell is hypromellose, and its claims include gluten free and lactose free. The valerian capsule uses a gelatin shell, and non-GMO is the only claim it prints. If you avoid gelatin, that decides it. Also count the 5-HTP capsule's 450 mg of calcium toward any other calcium you take.

When don't you need 5-HTP or an L-tryptophan supplement?

You probably don't need either if you eat protein through the day, take a medicine that affects serotonin, or already use a product that lists one.

  • You eat a varied diet with protein in it. Tryptophan is essential, but it arrives with the protein you already eat.
  • You take a medicine that affects serotonin, prescription or over-the-counter. Skip both unless your prescriber says otherwise.

Can you take 5-HTP and L-tryptophan together?

Taking 5-HTP and L-tryptophan together isn't something we'd do without a healthcare provider's approval. They're back-to-back steps in the same chain toward serotonin, so combining them stacks inputs neither label planned for. Our 10 mg melatonin capsule already pairs them in its blend, along with St. John's wort. If you take a medicine that affects serotonin, skip the combination entirely.

Is L-tryptophan the same as tryptophan?

L-tryptophan and tryptophan usually mean the same ingredient on a supplement label. Amino acids come in two mirror-image forms, called L and D, and the L form is the one in the proteins you eat. Labels add the "L-" to show which form is inside. MedlinePlus simply says tryptophan in its list of essential amino acids.

Is 5-HTP a stronger form of L-tryptophan?

5-HTP isn't a stronger form of L-tryptophan. It's a different compound, one step further along the same chain, according to MedlinePlus Genetics. Because tryptophan also builds protein and helps make niacin, milligrams of one don't convert into milligrams of the other. Judge each product against its own label.

Does eating turkey give you 5-HTP?

Turkey gives you tryptophan, not a measured dose of 5-HTP. The NIH Office of Dietary Supplements names turkey as high in tryptophan and notes your liver turns some of it into niacin. Your body can also make 5-HTP from tryptophan, but no food label shows how much of a meal takes that route.

How many days does one bottle of each cover?
